This is the key detail many people miss. The mission is not bugged, and nothing is secretly locked. The game simply expects you to follow those glowing trails to find each new group of Nodes linked to the Nest.

What To Do If Someone Destroyed The First Nodes And Left
Sometimes another teammate might destroy the first batch of Nodes and then disconnect or leave the game.
When that happens, you might join or continue the mission at the point where only the later Nodes remain. This can make things even more confusing because you never saw the first cluster and have no idea what you should be looking for.
In that situation, treat every glowing weak point you see as a potential Node connected to the Nest objective. If you reach a new area and the game still says you must destroy the Nest, start scanning for glowing Nodes on walls, ceilings, or around any corrupted-looking growths. Once again, follow the glowing vines until you reach the next patch of Nodes. Destroy each one and keep moving along the trail. Even if someone already took out earlier clusters, the remaining Nodes still work the same way, and the glowing trails will still guide you.

When the objective is not clear, it can be tempting to think something is broken and try desperate moves, such as jumping to your death to reset the area. That is not necessary for nest missions in Back 4 Blood.
