Finding Desmodus in ARK: Survival Ascended can be difficult on Lost Colony because the creatures stay high on cave ceilings, and some spawn areas are packed with hostile creatures. There are two reliable locations to check, with one being much safer for getting your first Desmodus. Bringing Blood Packs, a Parachute or Glider Suit, and a Crossbow with Grappling Hooks makes the tame much safer.
Find Desmodus at 82.7, 75.1
The easier Desmodus location is around 82.7, 75.1. Look for the blue building-like marker on the Lost Colony map and travel to the structure at this position.
Enter the cavern through the building. The cave itself is relatively small, but the ceiling is extremely high. Look upward to find Desmodus hanging from the roof. Only a few may spawn at once, so you will have fewer levels to choose from, but the location is easier to manage than the larger cave.

A Grappling Hook is particularly useful here because it lets you gain height and move closer to a Desmodus without relying on the creature to notice you from the cave floor.
Find the Large Desmodus Cave at 37.8, 20.3
The second location is around 37.8, 20.3. Look for a cave entrance surrounded by distinctive pink crystals, with a small lake below the area.

Follow the cave deeper until it opens into a large cavern near a waterfall. This chamber can contain a much larger number of Desmodus, giving you more opportunities to find a high-level one.
The tradeoff is danger. The route contains hostile insects, spiders, and other cave creatures. Attacking one Desmodus can also attract several others, so avoid provoking the entire group unless you are prepared to fight or escape.
How to Tame Desmodus
For a first tame, return to the cave at 82.7, 75.1 because dealing with a smaller number of Desmodus is safer.
Place plenty of Blood Packs in your inventory and approach close enough for a wild Desmodus to become aggressive. Using a Crossbow and Grappling Hook can help you reach a position where it notices you.

Allow the Desmodus to grab you. It will consume Blood Packs directly from your inventory while carrying you, gradually increasing its taming progress. Desmodus uses a passive taming method, so knocking it unconscious is not required.
If your Blood Packs run out while it is feeding, it can begin draining your Health instead. Keep extra Blood Packs available and carry a Parachute or Glider Suit because the Desmodus can release you high above the ground after the tame finishes.
The safest way to get your first Desmodus in Lost Colony is to visit 82.7, 75.1 with plenty of Blood Packs and fall-damage protection, while the cave at 37.8, 20.3 is better when you want more Desmodus levels to choose from.
